Effects of Adipose-Derived Mesenchymal Stem Cells and Human Amniotic Membrane on Sciatic Nerve Repair in Rats

چکیده
Background: Peripheral nerve injuries remain a great challenge for microsurgery despite the significant progress in recent decades. The current gold standard is autogenous grafting with success rate as low 50% long gaps. Current studies have focused on finding alternative methods bridging defects. Previous data demonstrated role of human amniotic membrane stimulating neural regeneration. On other hand, adipose-derived mesenchymal stem cells can differentiate into all three germ layers and could support repair. purpose this study was to compare without adipose tissue sciatic injury gap rats. Objectives: We aimed evaluate effectiveness Methods: Twenty-four male Wistar rats four random groups were used our study. In first group, repaired using inverse resected segment (Control group), second group (AM third an amnion sheet seeded (AM/ADMSCs last not repaired, both stumps sutured muscles. Results: All animals underwent procedures survived complication. function index hot plate test results significantly improved AM AM/ADMSCs compared Control (as care) (P>0.05). Based histopathology findings, regenerative fibers seen implanted area groups; however, surrounded by fibrosis (scar formation) group. axon count higher than experimental (P < 0.01). Conclusions: Our showed promotion regeneration gap, but adding only has no extra benefits, also causes more scar.

Comparison of Characteristics of Human Amniotic Membrane and Human Adipose Tissue Derived Mesenchymal Stem Cells
BACKGROUND Mesenchymal stem cells (MSCs) are ideal candidates for treatment of diseases. Amniotic membranes are an inexpensive source of MSCs (AM-MSC) without any donor site morbidity in cell therapy. Adipose tissue derived stem cells (ASCs) are also suitable cells for cell therapy. There is discrepancy in CD271 expression among MSCs from different sources. In this study, the characteristics...

متن کاملImmunomodulatory effects of human amniotic membrane-derived mesenchymal stem cells
Human amniotic membrane-derived mesenchymal stem cells (hAM-MSCs) are capable of differentiating into several lineages and possess immunomodulatory properties. In this study, we investigated the soluble factor-mediated immunomodulatory effects of hAM-MSCs. Mitogen-induced peripheral blood mononuclear cell (PBMC) proliferation was suppressed by hAM-MSCs in a dose-dependent manner as well as hAM-...
متن کاملEffects of gamma radiation on adipose-derived mesenchymal stem cells of human breast tissue
Background: During radiation therapy, stromal cells surrounding the tumor (e.g mesenchymal stem cells) may affect the treatment outcomes. We aimed to investigate the effects of gamma radiation on the mRNA expression of cytokines, DNA damage and population doubling time (PDT) of adipose-derived mesenchymal stem cells (ASCs). Material and methods: ASCs were enzymatically extracted from breast tis...
متن کاملEvaluation of the Effects of Adipose Derived Mesenchymal Stem Cells Cultured on Decellularized Amniotic Membrane in Wound Healing of Distal Part of the Limbs in Horse
Background: Equine wounds, particularly those involving the distal portion of the limbs, often undergo prolonged complex healing and may enter a non-healing state with obvious financial and welfare implications. Poor blood supply and lower levels of oxygen to the lower limbs lead to imbalance of growth factors related to wound healing. Decellular amniotic membrane has been widely used in soft t...
